Studies on the chemical constituents of the seeds of Clausena lansium and their anti-obesity activity

English Abstract

Clausena lansium (Rutaceae), called wampee, is a species of strongly scented evergreen tree 3–8 m tall, native to southeast Asia. C. lansium is known to abound with carbazoles and coumarins, which showed a variety of pharmacologic effects, such as anti-gastrointestinal inflammation, hepatoprotective, anti-tumor, etc. In order to support the research work, related literatures, including structure types and bioactivities of the components from C. lansium, were fully reviewed by this thesis. The phytochemical investigation was carried out on the 95% EtOH extract of the seeds of C. lansium. A total of 24 compounds were isolated with comprehensive chromatography technologies and identified with various spectroscopic methods. Among them, four compounds were new, including N-methyl-N-(2R/S)-acetyloxy-2- phenylethyl-3-phenyl-(2Z)-propenamide (1/2), N-methyl-N-(2-oxo-2-phenylethyl)-3- phenyl-(2E)-propenamide (3) and ο-clausenamide (4). The structures of 20 known compounds were identified by 1D-NMR and MS data. The seeds of C. lansium are used to treat acute and chronic gastrointestinal inflammation. Using high fat diet induced obese mice model, the water fraction of the 95% EtOH extract of the seeds from C. lansium was found to reduce body weight and fat mass, improve lipid profile and prevent insulin resistance. Further studies showed the water fraction can regulate gut microbiota towards a healthy balance. The above results indicated that the water fraction of the C. lansium seeds has anti-obesity activity through regulating intestinal flora.

芸香科(Rutaceae)黄皮屬植物黄皮(Clausena lansium)是一種常綠喬木,具有 濃郁的芳香氣味,廣泛分佈在東南亞地區。前期的研究表明,黃皮中主要化學成 分為生物碱、香豆素以及萜類成分,古籍記載,黃皮的葉子、根以及種子均可入 藥,其中黃皮種子口服可用於治療慢性和急性腸道炎症,現代藥理表明黃皮中咔 唑生物碱,香豆素等成分具有抗炎等生物活性。 本論文首先對黃皮屬植物的化學成分及藥理活性進行了系統的文獻綜述。採 用矽膠柱層析色譜、葡聚糖凝膠柱色譜(Sephadex LH-20)、ODS 及製備型高效液 相色譜(Prep-HPLC)等分離純化技術,對黃皮核乙醇提取物的化學成份進行了研 究,分離得到 24 個化合物,其中四個新化合物,命名為 N-methyl-N-(2R/S)- acetyloxy-2-phenylethyl-3-phenyl-(2Z)-propenamide (1/2), N-methyl-N-(2-oxo-2- phenylethyl)-3-phenyl-(2E)-propenamide (3) 和 ο-clausenamide (4)。通過一維和二 維的核磁共振波譜以及其他譜學技術鑒定了結構。另外,通過一維核磁共振波譜 和質譜鑒定了 20 個已知化合物的結構。 基於黃皮種子口服可用於治療慢急性腸道炎症,利用高脂食物誘導肥胖小鼠 模型,本研究發現黃皮核乙醇提取物的水部位可以有效減少小鼠體重和脂肪組織 重量,改善血脂,預防胰島素抵抗。進一步的研究表明,水部位可以明顯改善腸 道菌群,趨向更健康的比例。以上研究結果表明,黃皮 95%醇提水部位具有一定 的調節腸道菌群與抗肥胖作用。
