Chemical separation and analysis of Coreopsis tinctoria

English Abstract

Coreopsis tinctoria Nutt. (C. tinctoria) is an annual forb from the genus Coreopsis of Asteraceae family that is native in North America and now spreads worldwide. It is also known as snow chrysanthemum because it is distributed in alpine meadow above the altitude of 3000 m in Kunlun Mountain in China. Modern pharmacological researches have revealed that the extracts and components of C. tinctoria exhibit superior activities, including antioxidant, antidiabetic,etc. The major bioactive ingredients of C. tinctoria are flavonoids and phenylpropanoids. In order to develop and utilize C. tinctoria resources, systematic chemical studies and accurate quality control methods were necessary for investigation. The study provided references for the applications and quality evaluation of C. tinctoria. This dissertation includes four chapters. Chapter 1 reviewed and summarized the chemical constituents, pharmacological activities, quality evaluation methods of C. tinctoria. Chapter 2 consisted of chemical isolation and structural elucidation of compounds from flowers of C. tinctoria. Several isolation techniques, including silica gel, reversed-phase C18 column chromatography and preparative HPLC chromatography were used for the isolation and purification. 17 compounds were isolated and elucidated by means of UV-Vis, MS, 1H and 13C NMR, including 12 flavonoids, 2 phenylpropanoids and 3 coumaroylspermidines. Among them, 5 compounds were firstly isolated from the genus Coreopsis. In Chapter 3, screening of antioxidant components from C. tinctoria was investigated based on HPTLC-DPPH bioautography, and developed antioxidant bands on TLC plates were identified by injecting single standards. It is found that 5 flavonoids and 2 phenylpropanoids were the major antioxidants in C. tinctoria. This method could also serve as an anti-fake method for the discrimination of C. tinctoria and other cultivars. In Chapter 4, MAE and HPLC-DAD methods were established for quantitative analysis of 13 components in C. tinctoria. Chemometric analyses (HCA and PCA) on contents of compounds were investigated on different parts of C. tinctoria and other cultivars. The study provided reliable references for the quality control of C. tinctoria.
