A comparison study of five Lonicera species flower buds in China

English Abstract

Flos Lonicerae Japonicae (FLJ), the flower bud of Lonicera japonica Thunb, is a famous heat clearing and detoxifying herbal medicine. It was firstly recorded in “Ming Yi Bie Lu” in Han Dynasty, and used to treat fever, cough, headache and sore throat. According the Chinese Pharmacopoeia, it is an important drug in Chinese medicine with great bioactivity and wide range of use. And the Chinese Ministry of Health has approved that FLJ can be used as both food and medicine. However, there are several species of lonicera flower buds in local drug market and being used as herbal medicine as well. An effective method to ensure the safety and quality of FLJ is obviously urgent and necessary. In this study, five different species of lonicera flower buds, including lonicerae japonicae, were identified and evaluated by High Performance Thin Layer Chromatography (HPTLC), Gas Chromatography-Mass Spectrometer (GC-MS) and Fourier Transform Infrared Spectroscopy (FTIR) techniques, respectively. This thesis consists of three parts. Part 1 gave a preliminary review of FLJ. Firstly, there was a concise introduction FLJ. Secondly, the chemical constituents of FLJ were summarized. Additionally, the main pharmacological activities of FLJ were classified. And the ethnopharmacological use was present at last. In part 2, the comparison studies were conducted among those five lonicera flower buds using HPTLC, GC-MS and FTIR techniques. Part 3 was the results and conclusions. The results showed that HPLC and HPTCL are good preliminary steps to distinguish those flower buds. Through the GC-MS, the fat-soluble components mainly consist of hydrocarbons and fatty acids, and differences between those kinds of components were significant. Meanwhile, the FITR method analyzed the chemical composition of raw herb powder, which showed little differences between these species. Combined with HPLC, HPTLC, GC-MS and FITR methods and the clustering analysis, it is effective to identify different species of lonicera and thus provide fundamental basis to control the quality and evaluate the authenticity of FLJ.

Chinese Abstract

金銀花是忍冬科植物忍冬Lonicera japonica Thunb的花蕾或初開的花,亦 是有名的清熱解毒中草藥。它始載于漢代的《名醫別錄》一書中,并常用於治 療發燒、咳嗽、頭疼和咽喉腫痛。根據中國藥典記載,金銀花是一味具有顯著 活性和廣泛用途的中藥,中國衛生部也將其列為了藥食兩用藥材。然而,在許 多藥材市場上出現了其他品種忍冬花被當成藥材銷售的情況。因此建立一種有 效手段來確保金銀花藥材的質量安全是十分重要的。本課題選取了包括藥典標 準金銀花的五種忍冬科植物花蕾,分別使用了高效簿層色譜法(HPTLC)、氣相色 譜-質譜聯用法(GC-MS)和傅里葉紅外光譜法(FTIR)進行研究。 本課題由三個部分構成。第一部分對金銀花進行了初步的綜述:首先簡要 介紹了金銀花,其次總結了金銀花的主要化學成分,然後概括了金銀花的主要 藥理學活性,最後介紹了金銀花在民族藥理學中的作用。在第二部分中,我們 對五種忍冬花蕾使用了高效液相色譜法(HPLC)、高效簿層色譜法(HPTLC)、氣相 色譜-質譜儀聯用法(GC-MS)和傅里葉紅外光譜儀法(FTIR)進行比較學研究。第 三部分則主要對實驗結果進行了分析。實驗結果顯示高效液相色譜法和簿層色 譜法是初步鑒定五種忍冬花蕾的好方法。而通過氣相色譜-質譜儀聯用法得知忍 冬的脂溶性成分主要為烃类與脂肪酸,而且不同種忍冬间脂溶性成分相差較大。 同時,傅里葉紅外光譜儀法顯示藥材粉末的化學成分相差并不太多。本課題通 過聯合高效簿層色譜法、氣相色譜-質譜儀聯用法和傅里葉紅外光譜儀法,建立 了一種有效的鑒別金銀花的方法,并為其質量控制和品質鑒定提供了一定的研 究基礎。
