青陽參的化學成分研究 / Chemical study on the root of Cynanchum otophyllum Schneid / Shen Dongyan.

English Abstract

Cynanchum otophyllum Schneid(Chinese name Qingyangshen), a plant of the genus Cynanchum L. (Family: Asclepiadaceae), mostly distributed in the southwest of China, such as Guizhou and Yunnan provinces. As a traditional medicinal plant, the dried root of C. otophyllum has been used clinically in these areas to treat rheumatism, lumbar muscle strain and epilepsy. Modern pharmacological studies have revealed that it has anti-epileptic effect, anti-depressant effect, immunoregulation, liver protection and many other functions. C21 steroidal glycosides were reported to be the main bioactive compounds of C. otophyllum. In order to supply the materials for further exploration of quality control and pharmacological activities, a systematic chemical study on this herb has been carried out. 10 compounds were isolated from the dried root of C. otophyllum. Based on their physical and chemical properties and the spectral datas, their structures were identified as: 1-(3-(3,6-dihydroxy-2-methylbenzoyl)-2,4-dihydroxyphenyl)ethanone (1), qingyangshengenin 3-O-β-D-thevetopyranosyl-(1→4)-β-D-cymaropyranosyl- (1→4)-β-D-digitoxopyranoside (2), qingyangshengenin 3-O-β-D-thevetopyranosyl -(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-cymaropyranoside (3), caudatin 3-O-β-Dglucopyranosyl-(1→4)-β-D-oleandropyranosyl-(1→4)-β-D-cymaropyransyl-(1→4)-β -D-digitoxopyranoside (4) , caudatin 3-O-β-D-glucopyranosyl-(1→4)-β-D-oleandropyranosyl-(1→4)-β-D-cymaropyransyl-(1→4)-β-D-cymaropyranoside (5), caudatin 3-O-β-D-glucopyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-oleandropyranosy l-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-cymaropyranoside (6), caudatin 3-O-β-Dglucopyranosyl-(1→4)-β-D-glucopyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β- D-oleandropyranosyl-(1→4)-β-D-digitoxopyranoside (7), caudatin 3-O-β-Dglucopyranosyl-(1→4)-β-D-glucopyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-oleandropyranosyl-(1→4)-β-D-cymaropyranoside (8), caudatin 3-O-β-D-glucopyranosyl- (1→4)-β-D-glucopyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-oleandropyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-cymaropyranoside (9), caudatin 3-O-β-D-glucopyranosyl-(1→4)-β-D-glucopyranosyl-(1→4)-β-D-oleandropyranosyl- (1→4)-β-D-oleandropyranosyl-(1→4)-β-D-cymaropyranosyl-(1→4)-β-D-cymaropyra noside (10). An HPLC method was developed for simultaneous determination of seven major C21 steroidal glycosides (2, 3, 5, 6, 7, 8 and 10) and the method was applied to quantitatively analyze the seven compounds in C. otophyllum.

Chinese Abstract

青陽參(Cynanchum otophyllum Schneid)系蘿藦科鵝絨藤屬植物,又名青羊 參、青洋參等,分佈在我國的貴州、雲南等西南部地區。其性微溫,味甘微苦, 民間常用於治療風濕骨痛,腰肌勞損,癲癇等癥。現代藥理研究表明,青陽參具 有抗癲癇、抗抑鬱、免疫調劑和肝臟保護等作用,而其主要有效成分為 C21 甾體 皂苷類物質。爲了進一步探索青陽參的化學成分,為品質控制和藥理研究提供物 質基礎,本文對青陽參進行了化學成分研究。 本文對青陽參粗提物的氯仿萃取部位所含的主要化學成分進行了系統的研 究,從中分離得到了 1 種苯乙酮類物質和 9 種 C21 甾體皂苷類物質。根據化合物 的理化性質和波譜數據將 10 種化合物的結構鑒定為:2,6,2′,5′-四羟基, 3-乙酰基, 6′-甲基二苯酮(1),青陽參苷元 3-O-β-D-黃夾吡喃糖基-(1→4)-β-D-磁麻吡喃糖 基-(1→4)-β-D-洋地黃毒吡喃糖苷(2),青陽參苷元 3-O-β-D-黃夾吡喃糖基 -(1→4)- β-D-磁麻吡喃糖基-(1→4)- β-D-磁麻吡喃糖苷(3),告達亭 3-O-β-D-葡萄 吡喃羰基-(1→4)-β-D-夾竹桃吡喃糖基-(1→4)-β-D-磁麻吡喃糖基-(1→4)-β-D-洋 地黄毒吡喃糖苷(4),告達亭 3-O-β-D-葡萄吡喃羰基-(1→4)-β-D-夾竹桃吡喃糖 基-(1→4)-β-D-磁麻吡喃糖基-(1→4)- β-D-磁麻吡喃糖苷(5),告達亭 3-O-β-D-葡 萄吡喃羰基-(1→4)-β-D-磁麻吡喃糖基-(1→4)-β-D-夾竹桃吡喃糖基-(1→4)-β-D- 磁麻吡喃糖基-(1→4)-β-D-磁麻吡喃糖苷(6),告達亭 3-O-β-D-葡萄吡喃羰基 -(1→4)-β-D-葡萄吡喃糖基-(1→4)-β-D-磁麻吡喃糖基-(1→4)-β-D-夾竹桃吡喃糖 基-(1→4)-β-D-洋地黃毒吡喃糖苷(7),告達亭 3-O-β-D-葡萄吡喃羰基-(1→4)-β-D- 葡 萄 吡 喃 糖 基 -(1→4)-β-D- 磁 麻 桃 吡 喃 糖 基 -(1→4)-β-D- 夾 竹桃吡喃糖基 -(1→4)-β-D-磁麻吡喃糖苷(8),告達亭 3-O-β-D-葡萄吡喃羰基-(1→4)-β-D-葡萄 吡喃糖基-(1→4)-β-D-磁麻吡喃糖基-(1→4)-β-D-夾竹桃吡喃糖基-(1→4)-β-D-磁 麻吡喃糖基-(1→4)-β-D-磁麻吡喃糖苷(9),告達亭 3-O-β-D-葡萄吡喃羰基 -(1→4)-β-D-葡萄吡喃糖基-(1→4)-β-D-夾竹桃吡喃糖基-(1→4)-β-D-夾竹桃吡喃 糖基-(1→4)-β-D-磁麻吡喃糖基-(1→4)-β-D-磁麻吡喃糖苷(10)。其中化合物 2、 3、4 和 10 為新化合物,化合物 7 為首次從青陽參中分離得到。此外,本文建立了同時對 7 種 C21 甾體皂苷成分(2、3、5、6、7、8 和 10) 的 HPLC-DAD 的定量分析方法,并用建立的分析方法測定了青陽參中 C21 甾體 皂苷成分的含量。
