霸王花的化學成分研究

English Abstract

A systematic chemical study on the tradional Chinese medicine Bawanghua, the dried flower of Hylocereus undatus (Haw.) Britt. et Rose, was carried out. Thirteen compounds were isolated from the flowers of H. undatus. Their structures were elucidated as kaempferol (1), quercetin (2), isorhamnetin (3), kaempferol 3-O-α-L-arabinopyranoside (4), kaempferol 3-O-β-D-glucopyranoside (5), quercetin 3-O-β-D-glucopyranoside (6), isorhamnetin 3-O-β-D-glucopyranoside (7), kaempferol 3-O-β-D-galactopyranoside (8), quercetin 3-O-β-D-galactopyranoside (9), quercetin 3-O-β-D-rutinoside (10), kaempferol 3-O-β-D-rutinoside (11), kaempferol 3-O-α-L-rhamnopyranosyl-(1→6)-β-D-galactopyranoside (12), and isorhamnetin 3-O-α-L-rhamnopyranosyl-(1→6)-β-D-galactopyranoside (13). All the compounds were isolated from this plant for the first time. Then an HPLC method was developed for simultaneous quantification of six major flavonoid glycosides (5, 7, 10, 11, 12 and 13)in Bawanghua, and was used to evaluate the quality of different parts of Bawanghua as well as commercial Bawanghua samples from difference locations. Moreover, a simple HPLC method using three aglycones (1, 2 and 3) as chemical markers was established for rapid ruitin quality control of Bawanghua samples. 霸王花為仙人掌科量天尺屬量天尺Hylocereus undatus (Haw.) Britt. et Rose的 花,其性味甘微寒,《嶺南采藥錄》謂其可止氣痛, 理痰火咳嗽。可適用於治肺 結核、支氣管炎、頸淋巴結核、胰腺炎等。霸王花為嶺南地區著名的藥食兩用原 料,但目前尚無其化學成分的研究報導,為探索其化學成分以為其品質控制和藥 理評價提供物質基礎,本論文對霸王花進行了系統的化學成分研究。 本課題對霸王花中所含主要化學成分黃酮類物質進行了系統研究,運用多種 色譜分離和純化手段,從霸王花乙醇提取物中分離得到了13個黃酮醇及其苷類化 合物。根據化合物的理化性質和波譜數據,其結構分別被鑒定為:山柰酚 (kaempferol,1),槲皮素(quercetin,2),異鼠李素 (isorhamnetin,3),山柰 酚3-O-α-L-阿拉伯糖苷(kaempferol 3-O-α-L-arabinopyranoside,4),山柰酚 3-O-β-D-葡萄糖苷(kaempferol 3-O-β-D-glucopyranoside,5),槲皮素3-O-β-D-葡 萄糖苷(quercetin 3-O-β-D-glucopyranoside,6),異鼠李素 3-O-β-D-葡萄糖苷 ( isorhamnetin 3-O-β-D-glucopyranoside , 7 ), 山 柰 酚 3-O-β-D- 半 乳 糖 苷 (kaempferol 3-O-β-D-galactopyranoside,8),槲皮素3-O-β-D-半乳糖苷(quercetin 3-O-β-D-galactopyranoside , 9 ), 山 柰 酚 3-O-β-D- 芸 香 糖 苷 ( kaempferol 3-O-β-D-rutinoside , 10 ), 異 鼠 李 素 3-O-β-D- 芸 香 糖 苷 ( isorhamnetin 3-O-β-D-rutinoside,11),山柰酚3-O-α-L-鼠李糖(- 1→6)-β-D-半乳糖苷(kaempferol 3-O-α-L-rhamnopyranosyl- ( 1→6 ) -β-D-galactopyranoside, 12 ) 和 異 鼠 李 素 3-O-α-L-鼠李糖-(1→6)-β-D-半乳糖苷(isorhamnetin 3-O-α-L-rhamnopyranosyl- (1→6)-β-D-galactopyranoside,13)。上述化合物均為首次從霸王花中分離得到。 此外,本論文還建立了同時對其中6種主要黃酮苷(5,7,10,11,12和13) 的HPLC定量分析方法,並用新建立的方法對不同地區市售霸王花和霸王花不同 部位進行了品質評價。在此基礎上,進一步建立了霸王花黃酮苷酸水解後產物(三 種苷元1,2和3)含量的HPLC法快速分析方法,作為霸王花簡易快速的品質控制 方法。 關鍵詞:霸王花;黃酮;定量分析;品質評價
