Chemical studies on two traditional Chinese medicines-Eclipta prostrata and Corydalis yanhusuo

English Abstract

In order to search the chemical foundation of bioactivity, develop quality control method and production process, chemical studies of two traditional chinese medicine Eclipta prostrata L. and Corydalis yanhusuo W. T. Wang were carried out. The dried whole plant of Eclipta prostrata L. is known as Mo-han-lian? in traditional Chinese medicines. It belongs to Asteraceae family. The main cultivating places of E. prostrata are Jiangsu, Jiangxi, Zhejiang and Guangdong province. It has been used for the treatment of haemorrhage, hepatitis and snake venom poisoning, et al.. In order to elucidate the chemical compositions in this Chinese materia medica, systematical isolation was carried out by using several chromatographic separation techniques. Finally, 11 compounds was isolated and purified, 6 of them was identified as Pomalic acid (EP-1), Echinocystic acid (EP-2), oleanolic acid (EP-3), luteolin-7-O-B-D-glucoside (EP-4), stigmasterol-3-O-B-D-glucopyranoside (EP-5), stigmasterol (EP-6). Among them, EP-1 was isolated from this plant for the first time. Corydalis yanhusuoW. T. Wang (family Papaveraceae), yan-hu-suo in Chinese, is a famous traditional Chinese medicine. It is used in China to relieve pain of chest, abdomen and dysmenorrhea. It was reported that the benzylisoquinoline alkaloids, which are the main chemical components in C. yanhusuo, had anti-tumor activities. Study on the chemical compositions in C. yanhusuo is helpful for its anti-tumoral activity studies. Therefore, in present study, we had systematically investigated on the chemical constituents of C. yanhusuo. 19 compounds had been isolated from the 70% EtOH extract of this plant. By spectroscopic methods as well as the physical and chemical properties of separated compounds, the structures of 17 compounds were identified as dihydrochelerythrine (CY-1), dehydrocorydaline (CY-2), tetrahydrocolumbamine (CY-3), berberine (CY-4), palmatine (CY-5), tetrahydropalmatine (CY-6), isocorybulbine (CY-7), corydaline (CY-8), tetrahydrocoptisine (CY-9), jarrorrbizine (CY-10), coptisine(CY-11), emodin (CY-12), physcion (CY-13), 3B-hydroxy-olean-11,13 (18)-dien-28-oic acid (CY-14), stigmasterol (CY-15), 6-sitosterol (CY-16) and daucosterol (CY-17). Among them, CY-1 and CY-12~CY-14 were isolated from this plant for the first time.

震尋找傅統中藥墨旱蓮及延胡索生物活性的物質基儊,正碓進行品質評價,設計生產工藝等,我們對中藥墨早蓮及延胡素的化學成分進行了系統研究。 黑旱蓮叉稱旱蓮草、旱蓮子,爲菊科植物鰭腸(Eclipia prostrata L.)的全草,現代臨床硏究表明墨旱蓮對囚肝腎陰虛和血熱引|起的各種出血症都有很好的止血作用,對血熱鬚髮早白也有一定的療效。墨早蓮在我國主產於江蘇、江陋、浙江、廣東等地。國內外近年來對墨旱蓮的研究較爲活躍,爲了尋找新的活性化合 物,本課題對墨旱蓮的化學成分進行了系統研究。運用多種層析分離手段,現已 從墨旱蓮醇提取物中分離得到11個化合物,根據理化和波譜資料鑒定了其中6個 化合物的結構,分別爲坡模酸(BP-1),刺囊酸(EP-2),齊墩果酸(EP-3),木犀草 素-7-0-8-D-葡萄糖苷(P-4),豆甾醇-3-0-8-D-葡萄糖苷(EP-5),豆甾醇(P-6)。 其中坡模酸(EP-1爲從該植物首次分離得到。 延胡索又稱元胡,爲罌粟科植物延胡索(Corydalis yanhusuo W.T. Wang)的乾 燥塊莖,具有活血、利氣、止痛之功效,臨床用於治療胸脅、脘腹疼痛、經閉痛 經、產後瘀阻、跌打腫痛等症。延胡索化學成分較爲複雜,主要成分爲苄基異喹 啉類生物鹼。近年來有關於該類型生物鹼的抗腫瘤活性研究。但對於延胡索化學 基礎方面研究很少,爲了尋找延胡索中抗腫瘤有效成分,本課題對延胡索的化學 成分進行了系統研究。運用多種層析分離手段,從延胡索 70%乙醇提取物中分離 得到19 個化合物,根據理化和波譜資料鑒定了其中17 個化合物的結構,分别爲 二氫白屈菜紅城(CY-1)、去氫紫革城(CY-2)、四氫非洲防己胺(CY-3)、小檗㙎 (CY-4)、巴馬汀(CY5)、四氨巴馬汀(CY-6)、異紫革球城(CY-刀、紫堇城(CY-8)、 四氫黄連城(CY-9)、藥根城(CY-10),黄連城(C¥-11)、大黄素(CY-12)、大黄素甲(CY-13)、3B-羥基-旁墩果烷-11,13(18)-二烯-28-酸(CY-14)、豆甾醇(C¥-15)、B-穀甾醇(C¥-16和胡蘿蔔苷(C¥-17)。其中,CY-1及 CY-12~C¥-14 爲首次從延胡 索中分離得到
