利用毛細管電泳技術測定淫羊藿中4種黃酮類成分的含量

English Abstract

Modern analytical techniques are playing important roles in quality control of traditional Chinese medicine (TCM). A great progress on the research of TCM has been achieved by using modern analytical techniques including ultraviolet technology (UV), thin layer chromatography (TLC), high performance liquid chromatography (HPLC), gas chromatography-mass spectra (GC-MS), liquid chromatography-mass spectra (LC-MS), capillary electrophoresis (CE) and capillary electrophoresis-mass spectra (CE-MS). Epimedium, Yin-Yang-Huo, is a kind of traditional Chinese herbal medicine, which can nourish kidney, cure rheumatism and slower senescence. In this thesis, the first chapter is a review, which includes the research on Epimedium in recent years, especially the analytical methods used to determine the flavonoids in Epimedium, as well as the advance of CE and CE-MS. In the section of experiment, chemical compounds were isolated from Epimedium first. However, only little icariin and a red needle crystal, a non-flavonoid, were obtained. Then, the study was focused on developing a method for quantitative determining four flavonoids, including icariin, epimedin A, epimedin B and epimedin C, in Epimedium by using CE. Central composite design (CCD) and response surface methodology (RSM) were applied to optimize the conditions. The results showed that the resolution of all four flavonoids was more than 1.5 and well separated in 20 minutes at optimized condition, which suggested the developed method was simple and accurate. In addition, CE-MS was developed to analyze the four flavonoids in Epimedium. However, it was failed because of the limit of running buffer. At last, the parameters of pressurized solvent extraction (PSE) for extraction of flavonoids in Epimedium were optimized, which was also compared with ultrasonic extraction. The results indicated that the extraction efficiency had no significant difference between them, except the reproducibility of PSE was much better. Chinese Abstract

現代分析方法是中藥品質控制的重要組成部分。紫外測定(UV)、薄層色譜(TCL)、高效液相色譜(HPLC)、氣相色譜-質譜聯用(GC-MS)、液相色譜-質譜聯用(LC-MS)、毛細管電泳(CE)、毛細管電泳-質譜聯用(CE-MS)等一系列現代分析技術在中藥分析領域得到了充分的應用和發展,也使得中藥品質控制研究得到了迅速的發展。本文首先綜述了近年來國內外對中藥淫羊藿的研究進展,尤其是對淫羊藿中黃酮類成分的分析研究進展;還綜述了CE及CE-MS 技術的研究和應用進展。在論文實驗的第一部分,進行了淫羊藿中黃酮苷類成分化學標準品的製備工作,但成果有限,只得到了少量的淫羊藿苷粉末;還得到了1種紅色針狀結晶,但非黃酮類成分。實驗的第二部分是本論文的重點,建立了 CE 測定淫羊藿中4種黃酮苷含量的方法,測定的4種黃酮苷分別是羊 苷、朝 定A、朝 定B和朝 定C,均為淫羊藿中的主要活性成分。實驗採用星點設計-效應面優化法優化了毛細管電泳條件,使得4種黃酮苷的分離度均在1.5以上,且在20分鐘內得到分離。建立的測定方法簡便、快捷,精密度高、重現性好、回收率穩定,符合定量測定的要求。實驗的第三部分是CE-MS 測定淫羊 中4種黃酮苷含量的方法學研究,由於緩衝液種類的限制,最終未能使4種黃酮苷得到分離。但積累了一定的經驗,為下一步實驗奠定了基礎。最後一部分是關於加壓溶劑提取(PSE)和超聲提取法的比較。首先優化了PSE提取淫羊藿中黃酮類成分的各項提取參數,然後對比了PSE 和超聲提取,結果表明二者的提取效率無明顯差別,但PSE 的重現性明顯優於超聲提取。 關鍵字:毛細管電泳, 淫羊藿, 黃酮,加壓溶劑提取
