Abstract
Obective:To screen for and analyze the functions of the differentially expressed genes between abdominal aortic aneurysm and normal aortic tissue by using cDNA microarray technique. Methods:The total RNAs were respectively isolated from the normal aortic aneurysm, purified into mRNAs by Oligotex, and then were reverse-transcribed into cDNAs with the incorporations of fluorescent dUTP to make the hybridization probes. The mixed probes were hybridized to the cDNA microarray, which was scanned for the fluorescent signals and the differentially expressed genes between normal aortic and aortic aneurysm were determined and analyzed with GenePix Pro 3.0 software. Results:A total of 18 differentially expressed genes were detected (accounting for 0.42% of total genes), among which, 11genes were related to cell cycle and the other 7 related to cell apoptosis. The number of up regulated genes in aortic aneurysm is 9 (mean Ratio value: 3.860) and that down regulated is 9 (mean Ratio value: 0.294). Bio-informative analysis showned that these 18 genes might influence the growth and apoptosis of smooth muscle cells in abdominal aortic aneurysms. Conclusion:During the development of abdominal aortic aneurysms, the modulations of multigene expression would undergo various changes. There is a relationship of the cell cycle and apoptosis-related genes with the growth and apoptosis of smooth muscle cells in abdominal aortic aneurysms.
